目的:研究羟基喜树碱HCPT对人肝癌Hep G2细胞分化诱导作用的机制。方法:用免疫细胞化学染色检测增殖细胞核抗原表达;流式细胞仪检测细胞周期和野生型p53蛋白表达;端粒重复扩增法检测端粒酶活性。结果:以诱导分化剂量(5-20μg·L~(-1))的HCPT处理6 d,人肝癌Hep G2细胞明显受阻于G_2/M期,PCNA阳性表达率降低。经HCPT 5μg·L~(-1)处理6 d,Hep G2细胞的野生型p53蛋白表达明显增强,但端粒酶活性没有改变。结论:HCPT诱导人肝癌Hep G2细胞分化的作用与细胞阻滞于G_2/M期、PCNA表达降低及野生型p53蛋白表达增强有关。
Objective: To study the mechanism of differentiation induced by HCPT on human hepatoma Hep G2 cells. Methods: Immunocytochemical staining was used to detect the expression of proliferating cell nuclear antigen; Flow cytometry was used to detect the cell cycle and wild-type p53 protein expression; Telomere repeat amplification method was used to detect telomerase activity. RESULTS: After treatment with HCPT at different doses (5-20μg·L -1) for 6 days, human hepatoma Hep G2 cells were significantly arrested in G 2 /M phase, and the positive rate of PCNA expression was decreased. After treated with HCPT 5μg·L -1 for 6 days, the expression of wild-type p53 protein was significantly enhanced in Hep G2 cells, but the telomerase activity was not changed. Conclusion: The effect of HCPT on the differentiation of human hepatoma Hep G2 cells is related to the cell arrest in G 2 /M phase, the decrease of PCNA expression and the increase of wild-type p53 protein expression.
