目的:以总酚、总黄酮、多糖、槟榔碱、槟榔次碱和去甲基槟榔次碱4类药效组分为评价指标,基于紫外分光光度法、超高效液相色谱-二极管阵列检测法(UPLC-PDA),外观性状变化和水分含量检查联合探讨不同储藏环境和包装材料对槟榔质量的影响。方法:分别采用福林-酚法、三氯化铝显色法和苯酚-硫酸法显色,在775,510,489 nm波长下利用紫外分光光度法测定槟榔中总酚、总黄酮和多糖含量;三氯甲烷-甲醇(4∶1)超声提取,以BEH HILIC色谱柱(2.1 mm×100 mm,2.7μm),乙腈-10 mmol·L~(-1)乙酸铵水梯度洗脱,利用UPLC-PDA法在215 nm波长下定量3种指标性生物碱含量。结果:不同储藏环境和包装材料下贮藏1年,槟榔外观颜色加深,水分含量增加,上述4类药效组分含量均随储藏时间延长而降低,其中,高温高湿(P<0.01)和常温常湿(P<0.01),编织袋(P<0.01)和牛皮纸(P<0.01)因素影响显著,低温低湿和自封袋(P<0.05)储藏条件对药效组分的含量影响较小。结论:建议可将槟榔用自封袋包装储存在低温低湿环境中,以延长槟榔储藏时间,保证槟榔质量。
OBJECTIVE: To evaluate the pharmacodynamics of four kinds of total phenols, total flavonoids, polysaccharides, arecoline, arecoline and benacetyl subunit, based on UV spectrophotometry, ultra performance liquid chromatography-diode array (UPLC-PDA), changes in appearance and moisture content and water content inspection to explore the different storage environment and packaging materials on the quality of areca nut. Methods: The content of total phenols, total flavonoids and polysaccharides in Areca catechu were determined by Fulin-phenol method, aluminum trichloride colorimetric method and phenol-sulfuric acid method respectively at the wavelengths of 775, 510 and 489 nm. - methanol (4: 1), and eluted with BEH HILIC column (2.1 mm × 100 mm, 2.7 μm) and acetonitrile-10 mmol·L -1 ammonium acetate aqueous solution The content of 3 kinds of indicator alkaloids was quantified at 215 nm. Results: Under the different storage conditions and packaging materials for 1 year, the appearance color of betel nut was deepened and the moisture content increased. The content of the above four kinds of effective components decreased with the prolongation of storage time. High temperature and high humidity (P <0.01) and normal temperature (P <0.01), kraft paper (P <0.01) and Kraft paper (P <0.01). The storage conditions of low temperature and low humidity and ziplock bag (P <0.05) had little effect on the content of the effective components. Conclusion: It is suggested that betel nuts can be stored in ziplock bags in low temperature and low humidity environment to prolong the storage time of areca nuts and ensure the quality of areca nuts.
