目的探讨原癌基因蛋白质c-met核酶基因对胃癌细胞生物学行为的影响。方法将含c-met核酶基因的U1/met292质粒转染至胃癌肝转移的SGC-7901亚系L2细胞株,检测其对L2亚系细胞增殖能力、细胞周期、血管内皮生长因子(VEGF)、c-met蛋白及肝转移能力的影响。结果转染c-met核酶基因对细胞增殖能力、细胞周期没有明显影响,但同L2亚系相比,其VEGF、c-met蛋白表达明显下降(P<0.05),其肝转移能力明显减弱(P<0.05)。结论抑制胃癌细胞c-met的过度表达,将可能起到预防与治疗胃癌细胞肝转移的作用。
Objective To investigate the effect of protooncogene c-met ribozyme gene on the biological behavior of gastric cancer cells. Methods U1 / met292 plasmid containing c-met ribozyme gene was transfected into SGC-7901 sublineline L2 cell line with metastasis to gastric cancer. The cell proliferation, cell cycle, vascular endothelial growth factor (VEGF) , C-met protein and liver metastasis ability. Results Transfection of c-met ribozyme gene had no significant effect on cell proliferation and cell cycle. However, the expression of VEGF and c-met protein was significantly decreased compared with that of L2 subline (P <0.05) (P <0.05). Conclusion Inhibition of overexpression of c-met in gastric cancer cells may play a role in prevention and treatment of hepatic metastasis of gastric cancer cells.
