实验选用SD大鼠23只,分别在小脑前、后叶定位定量(10~15μl)注射单纯疱疹病毒I型(HSV1,HK-2株,滴度10-7)存活1~5d或10d后灌杀取脑切片,经抗HSV1血清行免疫组织化学ABC法反应。结果显示,免疫阳性神经元呈类似Golgi样染色,胞体和树突标记清晰。注射部位不同或术后动物存活时间不同,其标记细胞出现的数量和部位也不同。1.后叶注射后1d可出现小脑Purkinje细胞及小脑核的标记,术后2d标记细胞可进一步出现在脑桥核、下橄榄核、前庭内侧核等。术后3d则另可在中缝核群、网状结构、室周灰质、丘脑腹外侧核发现标记细胞。2.前叶注射后3d,除有与后叶相似结果外,标记神经元还出现在红核、楔核、楔形核、外侧网状核、巨细胞网状核和Cajal中介核。实验结果表明:HSV1(HK-2株)注入小脑后可在神经元内进行逆行和顺行输送,其在脑内的跨神经元输送主要是顺行的,输送距离与存活时间有关。在选择正确的存活时间和滴度的情况下,HSV1(HK-2株)是一种良好的用于中枢神经系统的神经束路追踪剂。如在小脑不同部位注射,可显示良好的小脑传出、传入神经元链的结构
Twenty-three Sprague-Dawley rats were used in this study. Survival of 1 ~ 5d or 10d post-injection of herpes simplex virus type I (HSV1, HK-2, titer 10-7) Killing brain slices, the anti-HSV1 serum immunohistochemistry ABC reaction. The results showed that immunopositive neurons were similar to Golgi-like staining, clear cell body and dendritic markers. Different injection site or postoperative animal survival time is different, the number and location of the labeled cells are also different. 1. After injection of the posterior lobe, the cerebellum Purkinje cells and cerebellar nuclei could be marked on the 1st day after injection. The labeled cells on the 2nd day after the injection of the posterior lobe could further appear in the pons, inferior olivary nucleus and medial vestibular nucleus. Three days later, another labeled cell could be found in the nucleus of the mesoma, reticular formation, periventricular gray matter and ventrolateral thalamic nucleus. 2. In addition to the similar results with the posterior lobe, the labeled neurons also appeared in the nucleus of red nucleus, wedge nucleus, wedge nucleus, lateral reticular nucleus, giant cell reticular nucleus and Cajal intermedia nucleus 3d after injection of the anterior lobe. The experimental results show that HSV1 (HK-2 strain) injected into the cerebellum can retrograde and anterogradely within the neurons. The cross-neuronal transport of HSV1 (HK-2 strain) in the brain is mainly anterograde. The transport distance is related to the survival time. HSV1 (HK-2 strain) is a good neural tracer for the central nervous system, choosing the right survival time and titer. If injected in different parts of the cerebellum, can show a good cerebellar transmission, afferent neuronal chain structure
