目的 :观察微囊化牛肾上腺嗜铬细胞 ( BCC)脑内移植对偏侧帕金森病 ( PD)样猴的行为和脑组织液中单胺类物质含量的影响 ;评价微胶囊的免疫隔离效果。方法 :以海藻酸钠 -多聚赖氨酸 -海藻酸钠 ( APA)为材料包裹BCC。右侧颈内动脉注射甲基 -苯基 -四氢吡啶 ( MPTP)诱发 PD样猴模型。将微囊化、非囊化 BCC或空微囊分别植入 PD样猴右侧尾状核和壳核 ,观察 PD样猴行为、尾状核及壳核微透析液中单胺类物质的变化。结果 :囊化组 ( n=6)和非囊化组 ( n=2 )移植后 1周开始 PD样猴左侧上肢活动明显增加 ,阿朴吗啡 ( Apo)诱发的异常旋转次数减少。非囊化组猴在移植 1~ 2个月后主动行为及 Apo诱发的异常旋转圈数逐渐退回到移植前水平 ;而移植囊化BCC6只 PD样猴的异常行为纠正效果 ,在观察的 7个月内仍存在。空囊组 ( n=1) PD样猴异常行为无变化。移植前 PD样猴右侧壳核、尾状核细胞外液中 DA、DOPAC和 HVA水平较左侧明显降低 ( P<0 .0 5 )。移植后 2、 7个月时 ,囊化组右侧壳核、尾状核细胞外液中 DA、DOPAC和 HVA水平较移植前明显提高 ( P<0 .0 5 ) ,但仍低于左侧的水平。结论 :PD样猴脑内移植 BCC后异常行为明显改善 ,微囊组的纠正效果明显优于非囊化组 ,其移植区 DA及其代谢产生明显增加。 APA微囊具有良好的免?
OBJECTIVE: To observe the effect of microencapsulated bovine adrenal chromaffin cells (BCC) intracerebral transplantation on the behavior of monophasic parkinsonism (PD) -like monkeys and the content of monoamines in brain tissue fluid. To evaluate the immunological isolation effect of microcapsules. Methods: BCC was coated with sodium alginate-polylysine-sodium alginate (APA). PD-like monkey model was induced by injecting methyl-phenyl-tetrahydropyridine (MPTP) into the right internal carotid artery. The microencapsulated, non-encapsulated BCC or empty microcapsules were implanted into the right caudate and putamen nuclei of PD-like monkeys. The changes of monoamines in PD-like monkeys, caudate and putamen microdialysates were observed . Results: In the first week after transplantation, the activity of the left upper limbs of PD monkey was significantly increased and the number of abnormal rotation induced by apomorphine (Apo) decreased in the non-encapsulated group (n = 6) and non-encapsulated group (n = 2). In the non-encapsulated group, the initiative behavior and the number of abnormal rotation induced by Apo gradually returned to the pre-transplantation level after 1 to 2 months of transplantation. However, abnormal behavior of the PDCC-like monkey in transplantation-encapsulated BCC6 was corrected. In the observed 7 Month still exists. There was no change in the abnormal behavior of PD monkey in the empty bag group (n = 1). The level of DA, DOPAC and HVA in the right putamen and caudate nucleus extracellular fluid of pre-transplantation PD mice were significantly lower than those of the left ones (P <0.05). At 2 and 7 months after transplantation, DA, DOPAC and HVA levels in the right putamen and caudate nucleus extracellular fluid of the encapsulated group were significantly higher than those before transplantation (P <0.05), but still lower than those of the left s level. Conclusion: The abnormal behaviors of PD-like monkey after transplantation of BCC were significantly improved. The correction effect of microcapsules group was better than that of non-encapsulated group, and the DA and its metabolism were significantly increased in the transplantation area.
