目的:研究神经管缺陷大鼠模型胚胎形态学改变及PLA2、COX-2表达。方法:16只SD雌性大鼠分为2组:第一组为正常对照组,8只,受孕后正常饲养;第二组为糖尿病模型组,8只,受孕后第6天经过尾静脉注射链脲佐菌素(STZ)构建先天性神经管缺陷组大鼠。妊娠12日后取出所有胚胎做HE染色观察胚胎形态学及免疫荧光染色的方法观察PLA2、COX-2表达。结果:(1)HE染色及免疫荧光发现糖尿病胚胎的神经管形态发育明显畸形(2)免疫荧光发现PLA2、COX-2糖尿病胚胎的表达明显降低。结论:PLA2、COX-2基因表达在SD大鼠神经管的发育过程中起着重要的作用。可能对临床治疗神经管畸形提供了一种新方法。
OBJECTIVE: To study morphological changes of embryonic neural tube defects and expression of PLA2 and COX-2 in rats with neural tube defects. Methods: Sixteen SD female rats were divided into two groups: the first group was normal control group and the other 8 were normalized after conception. The second group was diabetic model group, and the other 8 were injected with tail vein injection chain on the 6th day after conception Construction of congenital neural tube defect group with streptozotocin (STZ). All embryos were removed after 12 days of gestation. HE staining was used to observe embryonic morphology and immunofluorescence staining to observe the expression of PLA2 and COX-2. Results: (1) HE staining and immunofluorescence showed that neural tube morphology of diabetic embryos developed significantly deformity. (2) Immunofluorescence showed that the expression of PLA2 and COX-2 in diabetic embryos was significantly decreased. Conclusion: The expression of PLA2 and COX-2 plays an important role in the development of neural tube in SD rats. It may provide a new method for clinical treatment of neural tube defects.
