[Objective] To study on the antidepressant effect and mechanism of total flavonoids extracts from Apocynum venetum leaves (AV-extract). [Method] The LC-MS/MS method was applied to investigate the contents of monoamine neurotransmitters, such as norepinephrine (NE), dopamine (DA), and serotonin (5-HT), in rats brain tissues by chronic depression stress. [Result] The concentration of NE, DA and 5-HT in chronic stress depression rats’ brain decreased compared with the control group, while the contents of NE and DA in AV-extract group were higher than depression group, and the effect was more apparently with the increasing dosage of AV-extract. Treatment of 70 mg/kg AV-extract had the same effects with fluoxetine hydrochloride (3.5 mg/kg group). However, 5-HT content of the AV-extract group was close to the depression group, but the increased dosage of fluoxetine improved the content of DA, NE and 5-HT in the rat brain tissues of the depression group. [Conclusion] The study proved the neurotransmitter hypothesis of depression, and found out that AV-extract could reverse the decrease of NE and DA contents in rat brain tissues, which might be the antidepressant mechanism of A. venetum.
