目的 :探讨糖尿病肾病肾小球系膜细胞增殖、分泌功能和细胞内 [Ca2 + ]i改变及意义。方法 :链脲佐菌素(STZ)复制糖尿病模型 ,取肾进行肾小球系膜细胞培养 ,3 H -TdR法测定细胞增殖 ,ELISA法测定纤维连接蛋白 (FN)分泌量 ,用Fluro - 3探针经共聚焦显微镜检测 [Ca2 + ]i变化。结果 :糖尿病大鼠肾小球系膜细胞 ,3 H-TdR掺入率和FN分泌量较正常大鼠增加 ,基础 [Ca2 + ]i两组无差异 ,但糖尿病肾小球系膜细胞 [Ca2 + ]i对血管紧张素Ⅱ反应明显减弱。结论 :糖尿病肾小球系膜细胞增殖和分泌FN增加参与了糖尿病肾病细胞外基质的积聚 ,其 [Ca2 + ]i对AngII反应减弱可能参与了糖尿病肾病早期高滤过
Objective: To investigate the proliferation and secretion of glomerular mesangial cells in diabetic nephropathy and the significance of intracellular [Ca2 +] i change. Methods: Streptozotocin (STZ) was used to replicate the model of diabetes mellitus (DM). Kidney was used to culture mesangial cells. Cell proliferation was measured by 3 H - TdR method. FN secretion was measured by ELISA. Flur - 3 The probe was detected by confocal microscopy [Ca2] i change. Results: Compared with normal rats, the incorporation rate of 3H-TdR and the secretion of FN in mesangial cells of diabetic rats were increased. There was no difference in basic [Ca2 +] i between the two groups, but the expression of [Ca2 +] i response to angiotensin Ⅱ decreased significantly. CONCLUSION: The increased proliferation and secretion of FN in diabetic glomerular mesangial cells are involved in the accumulation of extracellular matrix in diabetic nephropathy. The decreased response of [Ca2 +] i to AngII may be involved in the early hyperfiltration of diabetic nephropathy
