目的:探讨人巨细胞病毒(HCMV)感染对人脐静脉内皮细胞(HUVEC)中ADAMTS13表达水平的影响。方法:通过培养原代HUVEC,建立HCMV感染HUVEC细胞模型。Western blot法检测HUVEC细胞中ADAMTS13蛋白表达水平的情况。结果:100PFU或更高毒力的HCMV可感染HUVEC细胞,导致明显的细胞病变:病毒感染后24h,细胞部分开始出现变圆,细胞内颗粒增多;48h后,部分细胞开始出现细胞膜破裂、细胞坏死;随着病毒感染时间的延长,细胞坏死比例逐渐增加。原代培养的HUVEC细胞裂解液及细胞上清中均有大量的ADAMTS13表达;与未感染HCMV细胞比较,感染HCMV48h后,HUVEC细胞裂解液及细胞上清中的ADAMTS13蛋白表达水平均显著下降,差异具有显著性(P<0.05)。结论:HCMV可感染HUVEC并通过抑制细胞中ADAMTS13的表达促进血栓形成。
Objective: To investigate the effect of human cytomegalovirus (HCMV) infection on the expression of ADAMTS13 in human umbilical vein endothelial cells (HUVECs). Methods: HUVEC cells infected with HCMV were established by culturing primary HUVECs. Western blot was used to detect the expression of ADAMTS13 in HUVECs. Results: HCMV infected with 100 PFU or higher could infect HUVEC cells, leading to obvious cytopathic effect. At 24 h after virus infection, the cytoplasm became more rounded and some intracellular granules increased. Conclusion: HCMV can infect HUVEC and promote thrombosis by inhibiting the expression of ADAMTS13 in cells.
