恶性肿瘤患者的生育能力由于受到放疗、化疗的影响而长期或永久丧失。睾丸干细胞移植可使他们的生育能力得到保存。对小鼠的自身或异体睾丸干细胞移植已见报道。本研究以睾丸体积比小鼠要大得多的反刍动物和灵长目动物为对象,旨在探索一种对肿瘤患者有效的生殖细胞移植方法。 离体实验材料包括50头公牛、20头小牛、4只猴(cynomolgus monkey)、4只大鼠的睾丸。6个人类睾丸由前列腺癌患者捐献。所有睾丸在注射前均冰冻保存。供体睾丸组织经剪切、连续酶切和离心,在注射前与台盼蓝染料混合。注射在超声波引导下进行。选择2~3个注射部位,注射体积为2~5ml,超声波频率为7.5MHZ,注射时间约30 min。 结果 成年公牛的睾丸网充满了注射液,在睾丸网交叉部位发现了成团的注射细胞。在注射部位附近的间质组织中也发现注
The fertility of patients with malignant tumors due to the impact of radiotherapy and chemotherapy and the long-term or permanent loss. Testicular stem cell transplantation can preserve their fertility. Mouse or allogeneic testicular stem cell transplantation has been reported. In this study, ruminants and primates whose testicular volume is much larger than that of mice are designed to explore a method of germ cell transplantation that is effective for cancer patients. In vitro experimental materials included 50 bulls, 20 calves, 4 cynomolgus monkeys, testes of 4 rats. Six human testes are donated by prostate cancer patients. All testicles were frozen prior to injection. Donor testis tissue is sheared, continuously digested and centrifuged, mixed with trypan blue dye prior to injection. Injection is performed under ultrasound guidance. Select 2 to 3 injection sites, injection volume of 2 ~ 5ml, ultrasonic frequency 7.5MHZ, injection time of about 30 min. Results Adult bull’s testicular mesh filled with injection found in the testicular cross-section of the injection of cells into the group. Note also found in interstitial tissue near the injection site
