目的研究褪黑素对高糖刺激下大鼠Müller细胞亚铁血红素氧化酶1(hemeoxygenase-1,HO-1)和抗氧化相关转录因子核因子相关因子2(nuclear factor erythroid 2-related factor 2,Nrf-2)表达的影响。方法体外培养和鉴定大鼠Müller细胞。将培养细胞分为正常糖对照组、甘露醇对照组、褪黑素加正常糖组、高糖组、褪黑素加高糖组及褪黑素加高糖加褪黑素膜受体阻滞剂Luzindole组。48 h后逆转录聚合酶链反应检测HO-1和Nrf-2的表达及变化;将培养细胞分为正常糖对照组、高糖组和甘露醇对照组,48 h后逆转录聚合酶链反应检测褪黑素膜受体的表达及变化。结果褪黑素加高糖组与高糖组相比,HO-1和Nrf-2表达明显增加(P<0.05)。褪黑素膜受体MT1和MT2在体外培养的大鼠Müller细胞中均有表达,且在高糖刺激下表达明显增加(P<0.05)。结论褪黑素可诱导高糖刺激下Müller细胞HO-1和Nrf-2表达增加,且为膜受体介导,褪黑素可能在糖尿病视网膜病变中起重要的抗氧化作用。
Objective To investigate the effect of melatonin on the expression of hemeoxygenase-1 (HO-1) and antioxidant factor-related transcription factor 2 (NF-κB) in rat Müller cells induced by high glucose , Nrf-2) expression. Methods Rat Müller cells were cultured and identified in vitro. The cultured cells were divided into normal glucose control group, mannitol control group, melatonin plus normal glucose group, high glucose group, melatonin plus high glucose group and melatonin plus high glucose plus melatoninergic membrane block Luzindole group. The expression and changes of HO-1 and Nrf-2 were detected by reverse transcription-polymerase chain reaction 48 h later. The cultured cells were divided into normal control group, high glucose group and mannitol control group. Results Compared with high glucose group, the expressions of HO-1 and Nrf-2 in melatonin plus high glucose group were significantly increased (P <0.05). Melatonin membrane receptors MT1 and MT2 were expressed in Müller cells cultured in vitro, and were significantly increased under high glucose (P <0.05). Conclusion Melatonin can induce high expression of HO-1 and Nrf-2 in Müller cells induced by high glucose, and is mediated by membrane receptors. Melatonin may play an important antioxidant role in diabetic retinopathy.
