本文报道在Martinek法(Martinek RG.CLin Chem1963;9(5):620)基础上改进的血清腺苷脱氨酶(ADA)比色测定法,并进行方法学探讨。 材料与方法 显色剂 1:液体酚(AR)11ml,亚硝基铁氰化钠(AR)50mg,加蒸馏水至200ml.显色剂 2:次氯酸钠溶液(CR)4ml,2.5％NaOH196ml,共200ml冰箱保存。15mMol/L腺苷底物液:腺苷(Chromat P)200mg加0.067Mol/L磷酸盐缓冲液(pH7.2)50ml。硫酸铵标准贮存液:无水硫酸铵(AP)0.1416g,加蒸馏水100ml。
This article reports the improved serum adenosine deaminase (ADA) colorimetric assay based on the Martinek’s method (Martinek RG. Chem Chem., 1963; 9 (5): 620) and the methodological study. Materials and Methods Reagent 1: 11ml of liquid phenol (AR), 50mg of nitroso-sodium ferricyanide (AR) and 200ml of distilled water Color reagent 2: 4ml of sodium hypochlorite solution (CR), 196ml of 2.5% Save the refrigerator. 15 mMol / L adenosine substrate solution: 200 mg of Chromat P plus 50 ml of 0.067 mol / L phosphate buffer (pH 7.2).
