采用SRAP分子标记技术分析我国8个川续断属药用种的遗传多样性和亲缘关系,应用POPGENE计算遗传参数,TREECONW软件进行聚类分析。结果表明:26对引物共检测到558条带,其中多态性条带539条;种间的PPB=96.59%,Na=1.965 9,Ne=1.337 5,H=0.214 3,I=0.342 3,表明8个川续断属药用种的遗传多样性较高;种内各遗传参数的平均值为PPB=6.97%,Na=1.069 7,Ne=1.031 1,H=0.018 7,I=0.029 1,表明种内遗传多样性较低;Nei’s基因多样性指数计算的遗传分化系数(Gst=0.912 6)说明大部分遗传变异存在于种间;聚类结果与传统经典分类的吻合度极高,且同种的不同植株均聚在一起,证实采用SRAP分子标记进行川续断属药用种的快速鉴别是可靠的,可作为川续断属植物亲缘关系研究的有效方法之一。
SRAP molecular markers were used to analyze the genetic diversity and genetic relationship of 8 medicinal species in China. POPGENE genetic parameters were calculated and TREECONW software was used for cluster analysis. The results showed that 558 bands were detected by 26 pairs of primers, of which 539 bands were polymorphic. Among them, PPB was 96.59%, Na = 1.965 9, Ne = 1.337 5, H = 0.214 3, I = 0.342 3, The results showed that the genetic diversity of the eight medicinal species of Chuanxuan is high. The average genetic parameters of each species were PPB = 6.97%, Na = 1.069 7, Ne = 1.031 1, H = 0.018 7, I = 0.029 1 , Indicating that the intraspecies genetic diversity is low. The genetic differentiation coefficient (Gst = 0.912 6) calculated by Nei’s gene diversity index indicates that most of the genetic variations exist in the interspecific species. The clustering results are in good agreement with the traditional classifications and The same species of different plants are clustered together, confirming the use of SRAP molecular markers for rapid identification of medicinal species Daphnia is a reliable, can be used as a genus Daphnia genus one of the effective methods of research.
