目的利用siRNA转染干扰沉默ATP结合盒式G型亚家族成员2(ABCG2)基因,并观察沉默前后T24恶性生物学行为的变化。方法 siRNA沉默ABCG2后,观察T24增殖、克隆形成、迁移、侵袭、对放化疗抵抗、成干细胞球能力的影响。结果 siRNA能够使ABCG2 mRNA和蛋白质的表达下降80%左右,ABCG2被下调后,T24的增殖能力、克隆形成能力、迁移和侵袭能力、对放化疗的抵抗能力、成干细胞球的能力均下降(P<0.05)。结论沉默ABCG2能够降低膀胱癌T24的恶性及干性,可以作为一个潜在的治疗靶点。
OBJECTIVE: To use siRNA transfection to interfere with the silencing of ATP binding cassette G-type subfamily member 2 (ABCG2) gene and to observe the change of malignant biological behavior of T24 before and after silencing. Methods After ABCG2 silencing siRNA, we observed the effects of T24 proliferation, colony formation, migration and invasion on the chemoresistance and chemosensitivity of stem cells. Results siRNA reduced the expression of ABCG2 mRNA and protein by about 80%. After ABCG2 was down-regulated, the proliferation, clonogenicity, migration and invasion of T24, the resistance to radiotherapy and chemotherapy, and the ability to differentiate into stem cells decreased <0.05). Conclusion Silencing ABCG2 can reduce the malignancy and dryness of bladder cancer T24, which may serve as a potential therapeutic target.
