[目的]证明马齿苋多糖对神经细胞的保护作用。[方法]用马齿苋水提多糖处理原代培养的胎鼠大脑皮层神经细胞,MTT法和PI-Hochest(33342)双染色法分析细胞成活率。[结果]马齿苋多糖能有效地提高谷氨酸和Aβ处理后的神经细胞相对成活率,马齿苋多糖对大脑神经细胞有促进作用,并呈剂量影响;Western blot证明了马齿苋多糖对小鼠大脑细胞保护作用的机理可能是阻碍了由谷氨酸诱导的细胞凋亡通路中P35向P25的裂解。[结论]马齿苋多糖对神经细胞有保护作用。
[Objective] To prove the protective effect of purslane polysaccharide on nerve cells. [Method] The primary cultured fetal rat cerebral cortical neurons were treated with polysaccharides of purslane by water extraction. The cell survival rate was analyzed by MTT assay and PI-Hochest (33342) double staining. [Result] The polysaccharide of Portulaca oleracea could effectively increase the relative survival rate of neurons after treatment with glutamic acid and Aβ. The polysaccharide of Portulaca oleracea promoted the neuronal activity of the brain and showed a dose-dependent effect. Western blot showed that polysaccharides of Portulaca oleracea The mechanism of protective effects on mouse brain cells may be hindered by the cleavage of P35 from P25 in glutamate-induced apoptotic pathways. [Conclusion] The polysaccharide of purslane has protective effects on nerve cells.
