目的检测肺鳞癌、肺腺癌细胞浆和细胞膜蛋白激酶Cα(PKCα)蛋白的表达,探讨PKCα在肺癌发生和发展中的作用。方法采用分别提取细胞浆和细胞膜蛋白,Westernblot检测37例鳞癌(鳞癌组)、23例腺癌(腺癌组)和17例肺良性病变的正常肺组织(对照组)中细胞浆、细胞膜PKCα蛋白表达含量,比较膜/浆PKCα比率。结果三组胞浆PKCα蛋白平均表达强度分别为0.0832±0.0152、0.1845±0.0089和0.1795±0.0150,三组间差异有统计学意义(P<0.01);鳞癌和腺癌组胞浆PKCα蛋白的平均强度显著高于对照组(P<0.01)。三组胞膜PKCα蛋白的平均强度分别为0.0331±0.0096、0.0918±0.0148和0.1009±0.0213,三组间差异有统计学意义(P<0.01);鳞癌和腺癌组织的胞膜PKCα蛋白的平均强度显著高于对照组(P<0.01)。三组膜/浆PKCα比值分别为0.3190±0.0172、0.5004±0.0950和0.5589±0.0865,三组间差异有统计学意义(P<0.01);鳞癌和腺癌组的膜/浆PKCα比值高于对照组(P<0.05)。结论肺鳞癌和腺癌细胞中无论是胞浆还是胞膜,PKCα蛋白的表达都增加,尤其是胞膜PKCα蛋白的表达增加幅度更大,提示PKCα不仅通过表达量增加参与肺鳞癌、肺腺癌的发生和发展,还以膜转位的活化方式参与细胞的癌性病变。
Objective To detect the expression of protein kinase Cα (PKCα) in lung squamous cell carcinoma and lung adenocarcinoma and to explore the role of PKCα in the development and progression of lung cancer.
