目的探讨瑞舒伐他汀对脑缺血再灌注损伤大鼠脑组织中E-选择素和L-选择素表达的影响。方法线栓法制作脑缺血再灌注模型,大鼠随机分为假手术组(sham组)、模型组(model组)和治疗组(test组),治疗组在模型制作前用瑞舒伐他汀5 mg·kg-1·d-1连续灌胃10 d。于缺血2 h再灌注,24 h后做神经功能缺陷评估,并断头取脑,应用免疫组织化学法、RT-PCR法和Western印迹法检测脑组织中E-选择素和L-选择素的表达。结果瑞舒伐他汀预处理能显著降低大鼠脑缺血再灌注损伤后神经功能缺陷评分及脑组织中E-选择素和L-选择素的表达(P<0.05)。结论瑞舒伐他汀可通过抑制E-选择素和L-选择素的表达从而发挥脑保护作用。
Objective To investigate the effect of rosuvastatin on the expression of E-selectin and L-selectin in brain tissue of rats with cerebral ischemia-reperfusion injury. Methods The cerebral ischemia-reperfusion model was made by the method of thread occlusion. The rats were randomly divided into sham group, model group and test group. The treatment group was treated with rosuvastatin 5 mg · kg-1 · d-1 for 10 days. The neurological deficits were assessed 24 hours after reperfusion at 24 hours after ischemia and brain was decapitated. Immunohistochemistry, RT-PCR and Western blotting were used to detect E-selectin and L-selectin expression. Results Rosuvastatin pretreatment significantly reduced neurological deficit score and E-selectin and L-selectin expression in rat brain after cerebral ischemia-reperfusion (P <0.05). Conclusion Rosuvastatin exerts cerebral protection by inhibiting the expression of E-selectin and L-selectin.
