采用纳米金(15 nm)标记神经元特异性烯醇化酶抗体(anti-NSE)获得金标记anti-NSE。在适宜的条件下,金标记anti-NSE可与神经元特异性烯醇化酶(NSE)发生免疫反应生成NSE的免疫金复合物,使纳米金发生凝集,该液相体系在620 nm处共振光散射信号显著增强。实验表明,NSE质量浓度在0.05~20 ng/m L范围内与共振光散射增强值呈现良好的线性关系,检出限为0.01 ng/m L,方法已用于人血清中NSE的定量分析。
Gold-labeled anti-NSE was obtained using nano-gold (15 nm) labeled neuron-specific enolase antibody (anti-NSE). Under appropriate conditions, the gold-labeled anti-NSE can immunoreactive with neuron-specific enolase (NSE) to generate NSE immunogold complexes that allow the gold nanoparticles to agglutinate. The liquid phase system resonates at 620 nm Scattering signal is significantly enhanced. The experimental results showed that the concentration of NSE showed good linearity with the enhancement of resonance light scattering in the range of 0.05 ~ 20 ng / m L with the detection limit of 0.01 ng / m L. The method has been applied to the quantitative analysis of NSE in human serum.
