骨桥蛋白(Osteopontin,OPN)是一种与免疫性疾病相关的多功能蛋白,是由被激活的巨噬细胞和淋巴细胞分泌的促炎细胞因子,在多种细胞中表达并通过与2种细胞黏附分子[整合素(α9β1、ανβ3、ανβ1、α4β1)和CD44]的结合而影响、调节许多生物学功能:包括免疫应答、组织重塑和血管增生。研究发现骨桥蛋白在炎症、癌症、细胞免疫和神经退行性疾病中明显升高。本文对骨桥蛋白在免疫相关性疾病中作用的分子机制作一概述,进而为OPN介导的相关疾病的治疗提供新的思路。
Osteopontin (OPN) is a multifunctional protein associated with immune diseases. It is a proinflammatory cytokine secreted by activated macrophages and lymphocytes and expressed in many kinds of cells. The binding of cell adhesion molecules [integrins (α9β1, ανβ3, ανβ1, α4β1) and CD44] regulates many biological functions including immune responses, tissue remodeling and angiogenesis. The study found that osteopontin significantly increased in inflammation, cancer, cellular immunity and neurodegenerative diseases. This article gives an overview of the molecular mechanism of osteopontin in immune-related diseases, and provides new ideas for the treatment of OPN-mediated diseases.
