目的研究西维美林对大鼠颌下腺和舌下腺放射治疗(放疗)后预防龋齿的作用。方法SD大鼠随机分为5组(n=10):阴性对照组(未放疗、未给药)、单纯给药组(未放疗)、放疗前给药组、放疗后给药组和阳性对照组(放疗+等体积生理盐水)。放疗前1d,大鼠口腔人工接种致龋的茸毛链球菌6715。放疗时,将大鼠颌下腺和舌下腺置于X-射线下,剂量为15Gy,剂量率2~3Gy·min-1,放疗1次;给药条件为:西维美林60mg·kg-1·d-1,ig,tid,于放疗前7d或放疗后当日开始给药。放疗后d28,按Keyes评分法评定大鼠平滑面龋齿和沟龋齿状况,并检测各组大鼠体重的改变情况。结果放疗前或后给予西维美林,大鼠平滑面龋齿和沟龋齿的评分均低于阳性对照组(P<0.05);放疗前给药组评分显著低于放疗后给药组(P<0.05)。放疗前或放疗后给予西美维林对大鼠体重的影响显著小于阳性对照组(P<0.05)。结论西维美林对放疗所致大鼠龋齿有预防作用,放疗前使用比放疗后使用效果好。
Objective To study the effect of simemeline on the prevention of tooth decay in rats after submandibular gland and sublingual gland irradiation (radiotherapy). Methods SD rats were randomly divided into 5 groups (n = 10): negative control group (no radiotherapy, no administration), simple therapy group (no radiotherapy), radiotherapy before treatment group, radiotherapy group and positive control Group (radiotherapy + equal volume of saline). One day before radiotherapy, the rat oral inoculated cariogenic Streptococcus sobrinus 6715. Radiotherapy, the rat submandibular gland and sublingual gland under the X-ray dose of 15Gy, the dose rate of 2 ~ 3Gy · min-1, radiotherapy once; dosing conditions: simethromycin 60mg · kg-1 · D-1, ig, tid, 7d before radiotherapy or on the day after radiotherapy. Radiotherapy after d28, according to Keyes score method to evaluate the smooth surface of caries and groove decayed tooth decay conditions, and test the changes of body weight in rats of each group. Results Before and after radiotherapy, the scores of carvedilol and ditch caries in the smooth surface of the rats were lower than those in the positive control group (P <0.05). The scores in the pre-radiotherapy group were significantly lower than those in the radiotherapy group (P < 0.05). Before and after radiotherapy, the effect of simeverin on the body weight of rats was significantly less than that of the positive control group (P <0.05). Conclusion Ximevermelin has a preventive effect on dental caries in rats induced by radiotherapy, and is better than radiotherapy before radiotherapy.
