[目的]建立一种NK细胞体外选择性高效扩增的实验体系,观察其体内的抗肿瘤活性。[方法]以干细胞培养基(SCGM)为培养基,在低剂量IL-2(100U/ml)联合14mer肽TKD(TKDNNLLGRFELSG,aa450-463)(2μg/ml)的培养条件下,从人外周血PBMC中高效扩增获得CD3-CD56+NK细胞,并建立肝癌裸鼠移植瘤模型,观察NK细胞在体内的抗肿瘤活性。[结果]建立了一种高效的从人外周血PBMC中选择性扩增NK细胞的体系,该NK细胞对表面表达HSP70的肝癌移植瘤模型具有显著的抗肿瘤作用。[结论]经过14mer肽TKD激活的NK细胞对表面表达HSP70的肿瘤具有很强的溶解活性。
[Objective] To establish an experimental system for the selective and efficient expansion of NK cells in vitro and observe the antitumor activity in vivo. [Method] With the culture medium of stem cell culture medium (SCGM) and human IL-2 (100U / ml) combined with 14mer peptide TKD (TKDNNLLGRFELSG, aa450-463) The CD3-CD56 + NK cells were efficiently amplified in PBMCs and the hepatocellular carcinoma xenografts in nude mice were established. The NK cells have a significant anti-tumor effect on the HCC xenograft model with HSP70 on the surface. [Conclusion] The NK cells activated by 14mer peptide TKD have strong lytic activity on tumors expressing HSP70 on the surface.
