已证实在细胞核中存在有甲状腺激素受体,对其生物学意义尚未弄清,为了阐明受体的生理作用,必先得到纯态受体蛋白的理化特征的资科。近曾报告受体的部分纯化,尚未能完全提纯,其困难之一是受体活性在储存中不稳定。作者对各种储存条件进行了研究,以表观缔合常数(Ka)与最大结合容量(Bmax)两个参数的乘积为指标,观察大鼠肝染色质在不同贮存条件下,其受体蛋白对T_3结合力的稳定性;同时也对受体蛋白经羟基磷灰石柱层析的纯化工作进行了研究。
It has been confirmed that there are thyroid hormone receptors in the nucleus, its biological significance has not yet been clarified. In order to clarify the physiological role of the receptor, we must first obtain the physical and chemical characteristics of the receptor protein. Recently, it has been reported that partial purification of the receptor has not yet been completely purified, and one of the difficulties is that receptor activity is not stable in storage. The authors studied various storage conditions and took the product of the apparent association constant (Ka) and the maximum binding capacity (Bmax) as indices to observe the changes of hepatic chromatin in rat liver under different storage conditions On the stability of T_3 binding force; also purified the receptor protein by hydroxyapatite column chromatography.
