目的:建立一种同时检测8种人参皂苷的HPLC检测方法,分析人参皂苷在人参全草各部位中的分布情况。方法:采用Discovery C18色谱柱(4.6 mm×250 mm,5μm),柱温35℃,流动相乙腈-水进行梯度洗脱,流速1 m L·min-1,检测波长203 nm,检测六年生人参的花蕾、叶、茎、芦头、支根、须根以及主根木栓层、韧皮部、木质部9个部位中人参皂苷Rg1,Re,Rf,Rb1,Rc,Rb2,Rb3,Rd的含量。结果:所建方法具有良好的重复性和溶液稳定性,8种成分可达到基线分离,并且具有较宽的线性范围和良好的线性关系,回收率在98%~102%。结论:首次对8种人参皂苷在人参全草中的分布进行了研究,所建方法可用于同时测定人参全草不同部位中8种人参皂苷的含量;8种人参皂苷在须根、花蕾中含量高,在茎中含量最低,主根中的人参皂苷则主要分布于木栓层。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of 8 ginsenosides and analyze the distribution of ginsenosides in all parts of the ginseng plant. METHODS: Using a Discovery C18 column (4.6 mm×250 mm, 5 μm), column temperature 35°C, mobile phase acetonitrile-water gradient elution, flow rate 1 m L·min-1, detection wavelength 203 nm, detection of six-year-old ginseng The contents of ginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, Rb3, Rd in the buds, leaves, stems, reed heads, branching roots, fibrous roots, and main root cork layer, phloem, and xylem parts. RESULTS: The established method has good repeatability and solution stability. The eight components can achieve baseline separation, and have a wide linear range and a good linear relationship. The recovery rate is 98% to 102%. Conclusion: The distribution of 8 ginsenosides in the whole ginseng plant was studied for the first time. The established method can be used to simultaneously determine the contents of 8 ginsenosides in different parts of the ginseng plant. The 8 ginsenosides are high in fibrous roots and buds. The content of ginseng saponins in the main root is mainly distributed in the cork layer.
