目的 探讨人工脂褐素的制备方法 ,及其组织化学特征和生物学效应。 方法 提取大鼠肝脏线粒体 ,氧化还原交联 ,制备人工脂褐素 ,小鼠腹腔内注射 ,做巨噬细胞吞噬实验。取 1~ 5d的腹腔巨噬细胞 ,作动态观察。 结果 1~ 5d的腹腔巨噬细胞多种组织化学染色和荧光示踪呈阳性反应 ,胞质内颗粒或荧光团明显 ,分布特征与阳性对照相似。 结论 人工脂褐素具有天然脂褐素的组织化学特征和生物学效应 ,可以作为替代品 ,用于抗衰老研究
Objective To study the preparation of artificial lipofuscin, its histochemical characteristics and biological effects. Methods Rat liver mitochondria were extracted and redox cross-linked to prepare artificial lipofuscin, and the mice were injected intraperitoneally for macrophage phagocytosis. Take 1 ~ 5d peritoneal macrophages for dynamic observation. Results 1 ~ 5d peritoneal macrophages by a variety of histochemical staining and fluorescent tracer positive reaction, intracytoplasmic particles or fluorophores obvious distribution characteristics similar to the positive control. Conclusion The artificial lipofuscin has the histochemical characteristics and biological effects of natural lipofuscin, which can be used as a substitute for anti-aging research
