【目的】探讨木薯品种西选03快繁技术,为其种苗规模化生产提供技术支持。【方法】以水培幼茎腋芽为外植体,调查MS基本培养基中添加不同用量6-BA和PP333对外植体芽诱导、增殖培养和生根的影响。【结果】外植体采用0.1%KMNO4预处理10 min后经75%酒精20 s+0.1%Hg Cl2(加吐温-80)5 min的灭菌效果较好,污染率为14.0%,成活率达79.3%。采用MS+6-BA 0.05 mg/L培养基进行初代诱导效果较好,外植体培养15 d腋芽萌发率达90.0%,且萌发芽生长旺盛,茎秆粗壮。无菌腋芽茎段在MS+6-BA 0.1 mg/L培养基中继代增殖效果最佳,芽增殖倍数为4.6。MS+PP3330.3 mg/L培养基诱导生根效果较好,生根率100.0%,培养30 d平均根数为6.9,叶色墨绿,植株健壮。【结论】以幼茎腋芽为外植体的快繁体系可用于西选03种苗规模化生产。
【Objective】 The purpose of this study was to explore the rapid propagation technology of cassava variety West selection 03 to provide technical support for the large-scale production of its seedlings. 【Method】 Axillary buds were used as explants to investigate the effects of different dosages of 6-BA and PP333 on root induction, proliferation and rooting of explants in MS basal medium. 【Result】 The explants were pretreated with 0.1% KMNO4 for 10 min and then sterilized by 75% ethanol for 20 s + 0.1% Hg Cl2 (plus Tween-80) for 5 min. The contamination rate was 14.0% and the survival rate Up to 79.3%. The initial induction with MS + 6-BA 0.05 mg / L medium was better. The germination rate of axillary buds reached 90.0% after 15 days of explant culture, and the germination buds grew vigorously and the stems were stout. The germination of axillary buds in MS + 6-BA 0.1 mg / L medium was the best, with multiplication ratio of 4.6. MS + PP3330.3 mg / L medium induced rooting is better, the rooting rate was 100.0%, the average number of roots cultured for 30 d 6.9, dark green leaves, plants robust. 【Conclusion】 The rapid propagation system using axillary buds of young stems as explants can be used for the large-scale production of 03 seedlings in Xizang.
