[目的]研究VNTR基因座D1S80在朝鲜族和汉族群体中的遗传多态性.[方法]采用PCR扩增结合聚丙烯酰胺凝胶电泳技术和银染法对207名无关朝鲜族个体和203名无关汉族个体的D1S80基因座进行群体遗传多态性调查.[结果]D1S80基因座在朝鲜族和汉族人群中共检出26个等位基因,共103种基因型,朝鲜族群体中检出25个等位基因,73种基因型;汉族群体中检出22个等位基因,71种基因型;两个群体皆符合Hardy-Weinberg平衡,朝鲜族群体的杂合度观察值、多态性信息量、非父排除率和个人识别能力分别为0.917 9,0.893 8,0.705 9,0.968 3,汉族群体中分别为0.803 0,0.870 3,0.684 2,0.968 8;将两个群体的等位基因分布频率分别与其他不同地区、民族、种族群体进行比较,结果证实D1S80基因座在不同地区、民族和种族之间存在着不同程度的差异.[结论]D1S80基因座在朝鲜族和汉族群体中具有高度多态性,可用于法医学个体识别、亲权鉴定和群体遗传学调查.
[Objective] The research aimed to study the genetic polymorphism of VNTR locus D1S80 in Korean and Han populations. [Methods] PCR amplification combined with polyacrylamide gel electrophoresis and silver staining were used to examine the genetic polymorphism of 207 unrelated Koreans and 203 Polymorphisms of D1S80 loci in unrelated Han individuals were investigated. [Results] A total of 26 alleles were detected in D1S80 locus in Korean and Han nationality, with a total of 103 genotypes and 25 Alleles and 73 genotypes; 22 alleles and 71 genotypes were detected in Han population; both populations were in accordance with the Hardy-Weinberg equilibrium, the observed heterozygosity, the amount of polymorphism information, The non-parent exclusion rate and personal identification ability were 0.917 9,0.893 8,0.705 9,0.968 3 respectively, and the Han population were 0.803 0,0.870 3,0.684 2,0.968 8 respectively. The allele distribution frequencies of the two groups were respectively Compared with other ethnic, ethnic and racial groups in different regions, the results showed that D1S80 locus was different in different regions, ethnicities and races. [Conclusion] The D1S80 locus is highly polymorphic in Korean and Han nationality Sex, can be used for forensic Individual identification, paternity testing and genetic investigation groups.
