目的 研究脑缺血再灌流后 b FGF基因表达的意义及其机制。方法 采用原位杂交和免疫组化的方法 ,观察大鼠脑缺血再灌流后 b FGF基因的表达及 MK- 80 1对它们的影响。结果 缺血 2 h再灌流 1h可见 b FGF表达增高 (P<0 .0 5 ) ,b FGF m RNA表达于 12 h达高峰 ,b FGF蛋白于 2 4h达高峰 ;MK- 80 1组与缺血组相比 ,于再灌流 6 h~ 48h b FGF表达减弱 (P<0 .0 5 )。结论 局灶性脑缺血再灌流可诱导 b FGF表达 ,MK- 80 1则能抑制脑缺血后 b FGF的表达。
Objective To investigate the significance and mechanism of bFGF gene expression after cerebral ischemia and reperfusion. Methods The in situ hybridization and immunohistochemistry were used to observe the expression of b FGF gene and the effect of MK-80 1 on them after cerebral ischemia and reperfusion in rats. Results The expression of b FGF increased at 2 h after reperfusion for 1 h (P <0.05). The expression of b FGF m RNA peaked at 12 h and b FGF protein peaked at 24 h. The expression of b FGF m RNA in ischemic group Compared with the control group, the expression of FGF was weakened at 6 h-48 h after reperfusion (P <0.05). Conclusion FGF can be induced by focal cerebral ischemia and reperfusion, and MK-80 1 can inhibit the expression of b FGF after cerebral ischemia.
