目的:研究银杏达莫注射液(Ginkgo leaf extract and dipyridamole injection,GLED)降低家兔动脉血压的作用及机制。方法 :采用颈总动脉插管法测动脉血压。经静脉累积注射GLED(0.35、0.7、1.4、2.8、5.6、11.2 mg/kg)观察血压的变化。并分为:左旋硝基精氨酸(10 mg/kg)、异搏定(10μg/kg)、普萘洛尔(0.5 mg/kg)、吲哚美辛(0.5 mg/kg)、缬沙坦(1.5 mg/kg)5组,观察它们对GLED(5.6 mg/kg)降压作用的影响。结果 :GLED能显著降低家兔动脉血压,并具有量效依赖性(收缩压:r=0.76,t=8.4,P<0.01;舒张压:r=0.76,t=6.5,P<0.01)。静脉注射10 mg/kg左旋硝基精氨酸后,GLED(5.6 mg/kg)的降压作用明显减弱,异搏定、普萘洛尔、吲哚美辛和缬沙坦对GLED(5.6 mg/kg)的降压作用无明显影响。结论 :GLED能显著降低家兔动脉血压,其机制可能与内皮释放的NO有关。
Objective: To study the effect and mechanism of Ginkgo leaf extract and dipyridamole injection (GLED) on reducing arterial blood pressure in rabbits. Methods: Arterial blood pressure was measured by carotid artery cannulation. After intravenous cumulative dose of GLED (0.35,0.7,1.4,2.8,5.6,11.2 mg / kg) observed changes in blood pressure. And were divided into three groups: L-NNA (10 mg / kg), verapamil (10 μg / kg), propranolol (0.5 mg / kg), indomethacin (0.5 mg / kg) (1.5 mg / kg), and their effects on the antihypertensive effect of GLED (5.6 mg / kg) were observed. Results: GLED significantly reduced arterial blood pressure in rabbits and was dose-dependent (systolic pressure: r = 0.76, t = 8.4, P <0.01; diastolic pressure: r = 0.76, t = 6.5, P <0.01). The antihypertensive effects of GLED (5.6 mg / kg) were significantly attenuated after intravenous injection of 10 mg / kg L-arnino-arginine, and the effect of verapamil, propranolol, indomethacin and valsartan on GLED (5.6 mg / kg) no significant effect on the antihypertensive effect. CONCLUSION: GLED can significantly reduce the arterial blood pressure in rabbits, and its mechanism may be related to the NO released by the endothelium.
