目的:研究二脱甲氧基姜黄素对体外培养人肝癌细胞HepG2 Caspase-3蛋白酶活性的作用。方法:MTT法检测二脱甲氧基姜黄素对体外培养人肝癌细胞HepG2的抑制作用;分光光度检测其对Caspase-3酶活性的影响;免疫组化法检测其对人肝癌细胞HepG2 Caspase-3蛋白的表达。结果:二脱甲氧基姜黄素对人肝癌细胞HepG2有明显的抑制作用(P<0.001)且呈剂量关系,与姜黄素有显著差异(P<0.001);二脱甲氧基姜黄素能增强Caspase-3酶活性,增强Caspase-3蛋白的表达。结论:二脱甲氧基姜黄素能激活Caspase-3蛋白活性,这可能也是它体外抑制肝癌细胞HepG2生长的机理之一。
Objective: To study the effect of didemethyloxy curcumin on HepG2 Caspase-3 protease activity in vitro. Methods: MTT assay was used to detect the inhibitory effect of didemethoxy curcumin on HepG2 cells cultured in vitro. The effect of didemethoxy curcumin on the activity of Caspase-3 was detected by spectrophotometry. The effect of didemethoxy curcumin on the activity of HepG2 and Caspase-3 Protein expression. Results: Didemethoxy curcumin had a significant inhibitory effect on HepG2 cells (P <0.001), and showed a dose-dependent relationship with curcumin (P <0.001). Dodeoxy-curcumin increased Caspase-3 activity, enhance the expression of Caspase-3 protein. CONCLUSION: Didemethoxy curcumin can activate the activity of Caspase-3 protein, which may be one of the mechanisms that it inhibits the growth of HepG2 cells in vitro.
