目的:研究紫杉醇对肿瘤细胞增殖及基质金属蛋白酶(MMP-2,MMP-9)表达的影响。方法:半定量RT-PCR法检测紫杉醇对体外培养的HO8910卵巢癌细胞MMP-2和MMP-9基因mRNA表达的作用。结果:(1)MTT法检测提示紫杉醇在浓度达100 nmol·L~(-1)以上时,对HO8910细胞生长抑制率>30%(P<0.001),并随浓度增加而升高(r=0.6574,P<0.01);(2)紫杉醇浓度在10 nmol·L~(-1)时MMP-2和MMP-9 mRNA表达降低(P<0.01),其改变呈剂量依赖性下降。结论:紫杉醇在抑制HO8910细胞增殖时,也抑制基质金属蛋白酶MMP-2和MMP-9mRNA的表达,因而,可能产生抑制肿瘤增殖和侵袭转移的作用。
Objective: To study the effects of paclitaxel on tumor cell proliferation and the expression of matrix metalloproteinase (MMP-2, MMP-9). Methods: The effect of paclitaxel on mRNA expression of MMP-2 and MMP-9 in HO8910 ovarian cancer cell line was detected by semi-quantitative RT-PCR. Results: (1) MTT assay showed that the growth inhibition rate of paclitaxel to HO8910 cells was higher than 30% (P <0.001) when the concentration was over 100 nmol·L -1 and increased with the increase of concentration (r = 0.6574, P <0.01). (2) The expression of MMP-2 and MMP-9 mRNA was decreased in a dose-dependent manner when paclitaxel concentration was 10 nmol·L -1 (P <0.01) Conclusions: Paclitaxel also inhibits the expression of matrix metalloproteinase-2 and MMP-9 mRNA while inhibiting the proliferation of HO8910 cells. Therefore, paclitaxel may inhibit the proliferation, invasion and metastasis of HO8910 cells.
