目的 采用原位移植法建立人乳腺癌免疫双缺陷鼠 SCID转移模型并研究移植瘤的生物学特性。方法 将人乳腺癌细胞系MDA MB 2 31瘤块原位移植于 6只SCID鼠乳房的脂肪垫上 ,用其癌转移淋巴结传代共传 1 8代 68只。采用光镜、电镜、免疫组织化学等手段对各代移植瘤的形态特征、移植瘤的侵袭和转移等生物学特性进行研究。结果 肿瘤移植成功率为 97% ,液氮冻存复苏移植成功率为 1 0 0 % ,荷瘤小鼠体内出现淋巴道转移 ,9代后转移率为 95 %。原位移植瘤病理组织学结果表明移植瘤的形态学特点与原始细胞系完全一致 ,而原代与晚代移植瘤之间的超微结构有差异 ,显示进一步恶化的特征。多指标免疫组织化学检测表明 ,原代与晚代移植瘤之间存在有癌基因及蛋白表达的差异。结论 该模型为人乳腺癌的研究的较理想模型
Objective To establish a SCID metastasis model of human breast cancer immune deficient mice and to study the biological characteristics of transplanted tumors by orthotopic transplantation. METHODS: Human breast cancer cell line MDA MB 2 31 was implanted orthotopically on the fat pad of the breasts of 6 SCID mice. Sixty-eight passages were passaged for their metastatic lymph nodes. Light microscopy, electron microscopy, and immunohistochemistry were used to study the morphological features of transplanted tumors and the invasion and metastasis of transplanted tumors. Results The success rate of tumor transplantation was 97%. The success rate of liquid nitrogen cryopreservation and transplantation was 100%. Lymph node metastasis occurred in tumor-bearing mice. The metastasis rate was 95% after 9 passages. The histopathological results of orthotopic transplantation tumors showed that the morphological characteristics of the transplanted tumors were completely consistent with the original cell lines, while the ultrastructure between the primary and late-generation transplanted tumors was different and showed further deterioration characteristics. Multiple-index immunohistochemistry showed that there were differences in the expression of oncogenes and proteins between primary and late-generation xenografts. Conclusion This model is an ideal model for the study of human breast cancer
