目的:对益气化瘀清热方进行拆方,观察各类含药血清对脂多糖(LPS)诱导下体外培养的大鼠肾小球系膜细胞(Ms C)表达IκB-αm RNA及其蛋白的影响,探讨不同中药的作用强度以及益气化瘀清热方治疗肾小球肾炎的作用机制。方法:采用RT-PCR法检测各组含药血清对Ms C表达IκB-αm RNA的影响,Wersten blot法检测各组含药血清对大鼠Ms C表达IκB-α蛋白的影响。结果:各组含药血清均能上调体外培养的Ms C表达IκB-αm RNA及蛋白,清热组作用大于化瘀组、益气组,其中益气组作用最弱,三者之间的差异均有统计学意义(P<0.05);与雷公藤多苷组比较复方组作用较强(P<0.05),清热组无显著性差异。结论:益气化瘀清热方及其拆方均能上调体外培养的大鼠Ms C表达IκB-αm RNA及其蛋白,复方组作用明显优于其他组,清热组、雷公藤多苷组次之,益气组最弱。
OBJECTIVE: To investigate the effect of various medicated serums on the expression of IκB-αm RNA and its protein in rat mesangial cells (Ms C) induced by lipopolysaccharide (LPS) in vitro, To explore the role of different Chinese herbal medicine and Yiqi Huayu Qingre Fang treatment of glomerulonephritis mechanism of action. Methods: RT-PCR was used to detect the expression of IκB-αm RNA in MsC group. Wersten blot was used to detect the effect of each serum group on MsC expression of IκB-α protein. Results: The drug-containing serum of each group could up-regulate the expression of IκB-αm RNA and protein in Ms C cultured in vitro. The effect of heat-clearing group was more than that of Huayu group and Yiqi group, while the effect of Yiqi group was the weakest. (P <0.05) .Compared with tripterygium glycosides group, the effect of compound group was stronger (P <0.05), while there was no significant difference between heat-clearing group. Conclusion: Yiqi Huayu Heat Decoction and its disassembled formulas can up-regulate the expression of IκB-αm RNA and its protein in Ms C of rats cultured in vitro. The effect of compound prescription is obviously better than that of other groups. Heat-clearing group and tripterygium glycosides group , Yiqi group the weakest.
