目的观察银杏黄酮对大鼠急性放射性海马损伤的影响。方法 156只SD大鼠随机分为空白对照组、模型组和银杏黄酮(GF)低、中、高剂量处理组。模型组和GF组予全脑12MeV电子线单次照射20Gy,GF干预后1、7、30d取材。HE染色观察病理变化,TUNEL法检测凋亡细胞数,Western blot检测B细胞淋巴瘤/白血病2(Bcl-2)和Bcl-2相关X蛋白(Bax),免疫组化检测Bax、Bcl-2、胶质原纤维蛋白(GFAP),Griess法检测一氧化氮(NO)。结果照射后脑细胞水肿,凋亡细胞数、凋亡相关基因Bax和Bcl-2、GFAP表达以及NO表达均增加。银杏黄酮干预后脑水肿减轻,细胞凋亡减少,GFAP表达下调,NO表达减少;GF干预后30d及GF高剂量组改变更明显。结论银杏黄酮对急性放射性海马损伤具有保护作用。
Objective To observe the effect of ginkgetin on acute radioactive hippocampal damage in rats. Methods 156 SD rats were randomly divided into blank control group, model group and ginkgo flavonoid (GF) low, medium and high dose treatment groups. The model group and GF group to the whole brain 12MeV electron beam single exposure 20Gy, 1,7,30 d after GF intervention drawn. The number of apoptotic cells was detected by TUNEL, Bcl-2 and Bax were detected by Western blot. The expressions of Bax, Bcl-2, Glial fibrillary protein (GFAP), Griess method for the detection of nitric oxide (NO). Results Brain edema, apoptotic cells, apoptosis related genes Bax and Bcl-2, GFAP expression and NO expression increased after irradiation. After ginkgetin intervention, brain edema was reduced, apoptosis was decreased, GFAP expression was decreased and NO expression was decreased; GFR 30d and GF high-dose group changed more obviously. Conclusion Ginkgetin has a protective effect on acute radiation-induced damage of hippocampus.
