目的:探讨晚期糖基化终产物受体(RAGE)、核转录因子-κB(NF-κB)基因表达与非缺血性糖尿病足发病的关系。方法:获取非缺血性糖尿病足患者及外伤截肢对照患者的足部全层皮肤组织各一块,采用实时PCR和Western蛋白质印迹的方法检测组织中RAGE、NF-κB的基因表达。结果:糖尿病足标本中可检测到RAGE和NF-κB的mRNA和蛋白质表达,其表达水平与对照组间差异有极显著统计学意义(P<0.01)。结论:RAGE、NF-κB介导的氧化应激及炎症反应是非缺血性糖尿病足发病中的重要病理过程。
Objective: To investigate the relationship between advanced glycation end-product receptor (RAGE), nuclear factor-κB (NF-κB) gene expression and non-ischemic diabetic foot disease. Methods: Whole foot skin tissues of non-ischemic diabetic foot patients and traumatic amputation control patients were obtained. Real-time PCR and Western blotting were used to detect the gene expression of RAGE and NF-κB. Results: The mRNA and protein expressions of RAGE and NF-κB were detected in diabetic foot specimens. The expression levels of RAGE and NF-κB were significantly different between the two groups (P <0.01). Conclusion: RAGE and NF-κB-mediated oxidative stress and inflammatory response are important pathological processes in non-ischemic diabetic foot disease.
