目的 观察雌激素对活性维生素D [1,2 5 (OH) 2 D3 ]调节人的类成骨细胞TE85作用的影响。方法 以TE85为成骨细胞模型 ,3 H 胸腺嘧啶参入法测细胞增殖 ,3 H 脯氨酸参入法测细胞胶原合成 ,放射免疫法测骨钙素(BGP)含量 ,对硝基苯酚法测碱性磷酸酶 (ALP)活性。结果 单独以 0 5~ 5nmol·L-11,2 5 (OH) 2 D3 处理细胞 96h ,3 H 胸腺嘧啶参入被抑制 ,作用呈剂量依赖性。 5nmol·L-11,2 5 (OH) 2 D3 可使细胞ALP活性增加 2 9 7%。雌二醇(E2 )可取消 1,2 5 (OH) 2 D3 对细胞增殖的抑制作用。两药合用时能促进细胞胶原、骨钙素及ALP的合成。结论 1,2 5 (OH) 2 D3 有促进成骨细胞骨形成的作用。 1,2 5 (OH) 2 D3 与E2 联合应用时 ,其促进成骨的作用增强
Objective To investigate the effect of estrogen on the activity of human osteoblasts TE85 activated by vitamin D [1,2 5 (OH) 2 D3]. Methods TE85 was used as a model of osteoblasts, 3 H thymidine incorporation method was used to measure cell proliferation, 3 H proline incorporation method was used to measure collagen synthesis and radioimmunoassay was used to measure the content of BGP. Phosphatase (ALP) activity. Results The cells treated with 0-5 ~ 5nmol·L-11,25 (OH) 2 D3 for 96h inhibited the 3H-thymidine incorporation in a dose-dependent manner. 5nmol·L-11,25 (OH) 2 D3 increased ALP activity by 297%. Estradiol (E2) abolished the inhibitory effect of 1,25 (OH) 2 D3 on cell proliferation. Combination of two drugs to promote cell collagen, osteocalcin and ALP synthesis. Conclusions 1,25 (OH) 2 D3 can promote osteoblast formation. 1,25 (OH) 2 D3 combined with E2, its role in promoting osteogenesis increased
