[目的]研究胰岛素样生长因子ⅡmRNA结合蛋白3(IMP3)在浸润性乳腺癌及其各分子亚型中的表达及其临床意义。[方法]收集189例浸润性乳腺癌组织标本,采用免疫组织化学法检测ER、PR和Her-2的表达情况,并据此将乳腺癌患者分为腺腔A型、腺腔B型、Her-2过表达型和基底细胞样型。用免疫组织化学法测定4种分子亚型乳腺癌组织中IMP3的表达情况,并分析IMP3表达与乳腺癌患者临床病理特征的关系。[结果]18.5%浸润性乳腺癌组织中有IMP3表达。IMP3的表达与组织学分级、Ki-67指数有关(P<0.01),组织学分级越高及Ki-67指数高者IMP3表达越高。IMP3在腺腔A型、腺腔B型、Her-2过表达型和基底细胞样型中表达率分别为5.3%、10.0%、21.4%和58.3%。Her-2过表达型的IMP3表达率高于腺腔A型,差异有统计学意义(P<0.05)。基底细胞样型表达率高于腺腔A型、腺腔B型及Her-2过表达型,差异有统计学意义(P<0.001)。IMP3阳性表达者生存率显著低于阴性表达者(P<0.001)。[结论]IMP3在基底细胞样型乳腺癌组织中高表达,其表达可作为乳腺癌预后的评估指标之一,并有可能成为基底细胞样型乳腺癌靶向治疗的一个新靶点。
[Objective] To investigate the expression and clinical significance of insulin-like growth factor Ⅱ (mRNA) binding protein 3 (IMP3) in invasive breast cancer and its molecular subtypes. [Method] A total of 189 cases of invasive breast cancer tissues were collected and the expression of ER, PR and Her-2 were detected by immunohistochemistry. Breast cancer patients were divided into adenocarcinoma type A, gland cavity type B, Her -2 overexpression and basal cell type. The expression of IMP3 in four molecular subtypes of breast cancer was detected by immunohistochemistry and the relationship between IMP3 expression and the clinicopathological features of breast cancer was analyzed. [Results] The expression of IMP3 in 18.5% of invasive breast cancer tissues. The expression of IMP3 was correlated with histological grade and Ki-67 index (P <0.01). The higher the histological grade and the higher Ki-67 index were, the higher IMP3 expression was. The expression rates of IMP3 were 5.3%, 10.0%, 21.4% and 58.3% in glandular cavity type A, glandular cavity type B, Her-2 overexpression and basal cell type respectively. The expression of IMP3 in Her-2 overexpression group was higher than that in adenocarcinoma type A group (P <0.05). The expression rate of basal cell was higher than that of glandular cavity A, glandular cavity B and Her-2 overexpression (P <0.001). The survival rate of IMP3 positive expression was significantly lower than that of negative expression (P <0.001). [Conclusion] IMP3 is overexpressed in basal-like breast cancer tissues, and its expression may be used as a prognostic indicator of breast cancer and may be a new target of targeted therapy in basal-like breast cancer.
