目的:优选知母治疗湿热痹症有效物质组纯化工艺参数。方法:以芒果苷为指标,HPLC测定含量,对8种不同类型树脂进行考察,优选最佳树脂型号及其纯化条件。结果:确定最佳纯化条件为HPD-400型大孔树脂,加2 BV提取液进行吸附,分别用3 BV水,6 BV 30%乙醇洗脱。纯化后芒果苷纯度比原药液中含量提高6.6倍,总固体物减少87.3%。结论:纯化后药液中多糖等杂质明显减少,以芒果苷为代表的皂苷类成分得以较好的保留,为知母治疗湿热痹证研究提供前期研究基础。
Objective: To optimize Zhimu treatment of diarrhea active substance group purification process parameters. Methods: Mangiferin as an index, determination of content by HPLC, eight kinds of different types of resin were investigated, the best choice of the best resin type and purification conditions. Results: The best purification conditions were identified as HPD-400 macroporous resin, and 2 BV extract was used for adsorption, respectively, with 3 BV water and 6 BV 30% ethanol. Purified mangiferin purity than the original liquid content increased by 6.6 times, a total reduction of 87.3% solids. CONCLUSION: Polysaccharides and other impurities in the medicinal solution are significantly reduced after purification. The saponin components represented by mangiferin are well preserved, providing preliminary research basis for the study of Zhimu treating damp-heat syndrome.
