鉴定冬麦越冬的抗寒能力有多种方法,本文介绍的是一种较为简便快速的方法——红四唑染色测定法。 一、原理 红四唑(化学名为2,3,5一氯化三苯基四氮唑,符号TTC)是氧化还原色素,配制成的水溶液为无色,当它侵入生活组织时,由于生活细胞里脱氢酶的作用,它作为受氢体,接受生活细胞里基质脱下的氢离子,被还原后逐渐变为不溶于水的三苯基甲(月朁)(红色)。这样,我们可以依据越冬麦苗分蘖节和生长锥,在一定温度下与红四唑作用显红色所需的时间和红色的深浅,来判断其生活力。
