目的蜂毒素对膀胱癌模型大鼠膀胱癌抑制作用,并探讨其可能的机制。方法 MNU制作大鼠膀胱癌原位模型,分别用不同浓度的蜂毒素行膀胱灌注,观察大鼠成活率,计算肿瘤抑制率,RT-PCR和Western blotting检测P16的mRNA和蛋白表达变化。结果蜂毒素对模型大鼠行膀胱灌注后,大鼠生存率为64.15%,肿瘤抑制率为24.03%,RT-PCR和Western blotting检测P16的mRNA和蛋白表达增加,呈剂量相关性。结论蜂毒素能够抑制膀胱癌模型大鼠肿瘤生长,其机制可能与提高P16表达有关。
Objective To investigate the inhibitory effect of melittin on bladder cancer in bladder cancer model rats and to explore its possible mechanism. Methods MNU was used to establish rat bladder cancer in situ model. The different concentrations of melittin were used to perform bladder perfusion. The survival rate of rats was observed, and the tumor inhibition rate was calculated. The mRNA and protein expression of P16 were detected by RT-PCR and Western blotting. Results After melittin was injected into the bladder in rats, the survival rate was 64.15% and the tumor inhibition rate was 24.03%. The mRNA and protein expression of P16 increased by RT-PCR and Western blotting in a dose-dependent manner. Conclusion Melittin can inhibit tumor growth in bladder cancer model rats, and its mechanism may be related to increasing P16 expression.
