植物蔗糖转运蛋白负责蔗糖的跨膜运输,在韧皮部介导的源-库蔗糖运输以及库组织的蔗糖供给中起关键作用.本研究利用RT-PCR方法,从玉米自交系吉853中获得位于3号染色体上蔗糖转运蛋白(SUT4)的基因,命名为ZmSUT4-J,测序结果表明开放阅读框(ORF) 1506 bp,编码501个氨基酸,利用生物信息学软件对其编码蛋白的理化性质、结构和功能进行了分析和预测;并借助荧光定量RT-PCR方法对ZmSUT4的组织表达特异性进行了分析.同时为了验证其生物学功能,构建了适合玉米遗传转化的表达载体,为进一步利用ZmSUT4基因开展玉米高产转基因育种的研究工作奠定了基础.“,”Sucrose transporters are responsible for trans-membrane translocation of sucrose,and play important roles on phloem-mediated source-sink sucrose transportation and supply sucrose to sink tissues.The ZmSUT4 gene was subcloned from Zea mays inbred line Ji 853,termed ZmSUT4-J.The sequence analysis revealed that ZmSUT4-J has an open reading frame (ORF) of 1506bp and encodes 501 amino acids residues.The predicted protein has 12 transmembrane domains according to the bioinformatics analysis.Using tissue expression pattern analysis,this gene was proved to be specifically expressed in young leaf.To elucidate biological functions of ZmSUT4,the expression vector for transformation to monocot plants was constructed.
