目的 对制备的可溶性抗HRP Ⅱ单链抗体进行初步应用研究。方法 对从抗恶性疟原虫红内期噬菌体抗体库中筛选到的能分泌特异性抗HRP Ⅱ单链抗体克隆株进行可溶性诱导表达 ,采用Dipstick 免疫胶体金模型 ,检测重组HRP Ⅱ和疟疾患者血样。结果 以该抗体包被的Dipstick层析条能特异性检测重组HRP Ⅱ抗原 ,并能与恶性疟患者血样中天然HRP Ⅱ抗原起阳性反应 ,最低检测抗原量为 1ng/ml,敏感度和特异度分别为 81 8%和 85 7% ,与抗HRP Ⅱ单抗 3A3相比差异无显著性 (P >0 0 5 )。结论 所研制的抗HRP Ⅱ单链抗体具有作诊断试剂的潜质。
OBJECTIVE To study the preliminary application of soluble anti-HRP II single-chain antibody. Methods The soluble secreted specific anti-HRP Ⅱ monoclonal antibody clone was screened from the red phage antibody library of Plasmodium falciparum. The blood samples of recombinant HRP Ⅱ and malaria patients were detected by Dipstick immunogold assay. Results The antibody was coated with Dipstick chromatography strip to detect specific recombinant HRP Ⅱ antigen and could react positively with native HRP Ⅱ antigen in blood samples of patients with P. falciparum. The lowest detection antigen level was 1 ng / ml. The sensitivity and specificity Respectively 81 8% and 85 7%. There was no significant difference between anti-HRP Ⅱ McAb and 3A3 (P> 0.05). Conclusion The anti-HRP Ⅱ single-chain antibody developed has the potential as a diagnostic reagent.
