目的探讨多发性骨髓瘤(MM)患者瘤细胞中第10号染色体磷酸酶和张力蛋白同源缺失性基因(PTEN)蛋白、血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)及转化生长因子β1(TGF-β1)的表达,分析其在疾病中的意义。方法用免疫组化SP法测定49例MM患者、30例对照及26例化疗有效患者PTEN、VEGF、bFGF及TGF-β1的表达,应用χ2检验分析。结果 MM患者PTEN及TGF-β1的表达显著低于对照组及化疗有效组(P<0.05),而VEGF、bFGF的表达显著高于对照组及化疗有效组(P<0.05);MMⅠ+Ⅱ期组VEGF、bFGF表达率显著低于Ⅲ期组(P<0.05),而Ⅰ+Ⅱ期组PTEN、TGF-β1显著高于Ⅲ期组(P<0.05)。结论 PTEN、VEGF、bFGF及TGF-β1的异常表达可能在疾病的发展中起一定的作用,三者联合检测有助于判断疾病分期及预后。
Objective To investigate the expression of PTEN protein, vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in tumor cells of patients with multiple myeloma (MM) And transforming growth factor-β1 (TGF-β1) expression, and analyze its significance in the disease. Methods The expressions of PTEN, VEGF, bFGF and TGF-β1 in 49 MM patients, 30 controls and 26 chemotherapy-effective patients were determined by immunohistochemical SP method. Results The expression of PTEN and TGF-β1 in MM patients was significantly lower than that in control group and chemotherapy group (P <0.05), while the expression of VEGF and bFGF was significantly higher than that in control group and chemotherapy group (P <0.05) The expression of VEGF and bFGF in group Ⅰ and Ⅱ was significantly lower than that in group Ⅲ (P <0.05), while the expression of PTEN and TGF-β1 in group Ⅰ + Ⅱ was significantly higher than that in group Ⅲ (P <0.05). Conclusion The abnormal expression of PTEN, VEGF, bFGF and TGF-β1 may play a role in the development of the disease. The combined detection of the three can help to determine the stage and prognosis of the disease.
