目的探讨急性冠脉综合征(ACS)患者外周血中CD4+CD25+调节性T细胞(Treg)的异常与疾病的关系。方法28例ACS患者(ACS组)2、6例稳定性心绞痛(SAP)患者(SAP组)和30例正常人(对照组)作为入选对象,均采用流式细胞术检测各组外周血中Treg表达百分比,并检测各组Treg的功能状态,ELISA法检测循环及上清液中细胞因子TNF-α和IL-10浓度,常规检测循环CRP及血脂水平。结果外周血中Treg表达百分比水平及功能ACS组显著低于SAP组(P<0.05)和对照组(P<0.05),CRP、TNF-α的水平ACS组显著高于SAP组(P<0.05)和对照组(P<0.05),ACS组IL-10的水平显著低于SAP组(P<0.05)和对照组(P<0.05)。结论ACS患者外周血中Treg比例减少及功能的降低可能破坏了外周免疫耐受的平衡,参与炎症反应激活致动脉粥样硬化发生发展这一病理过程。
Objective To investigate the relationship between the abnormalities of CD4 + CD25 + regulatory T cells (Tregs) and the disease in peripheral blood of patients with acute coronary syndrome (ACS). Methods Twenty-eight patients with ACS (ACS group), 2,6 patients with SAP (SAP group) and 30 normal subjects (control group) were enrolled in this study. Flow cytometry was used to detect Treg The percentage of expression of Treg in each group was detected. The levels of cytokines TNF-α and IL-10 in circulation and supernatant were detected by ELISA, and the levels of circulating CRP and blood lipid were detected routinely. Results The percentages of Treg in peripheral blood and functional ACS group were significantly lower than those in SAP group and control group (P <0.05). The levels of CRP and TNF-α in ACS group were significantly higher than those in SAP group (P <0.05) (P <0.05). The level of IL-10 in ACS group was significantly lower than that in SAP group (P <0.05) and control group (P <0.05). Conclusions The decrease of Treg proportion and the decrease of function in the peripheral blood of patients with ACS may undermine the balance of peripheral immune tolerance and participate in the pathological process of the development of atherosclerosis caused by inflammatory response.
