目的观察碘缺乏和甲状腺功能减退损伤内嗅皮质对钙调蛋白依赖性蛋白激酶(calcium/calmodulin pro-tein kinase II,CaMKⅡ)表达的影响。方法健康2月龄雌性Wistar大鼠,交配妊娠后,取孕鼠28只,按体重随机分成对照组、甲状腺功能减退组和碘缺乏组,甲状腺功能减退组根据饮水中含丙基硫尿嘧啶(propylthiouracil,PTU)剂量分为5ppm组和15 ppm组,每组7只孕鼠。分别于PN7、PN14、PN21、PN28和PN42时,每组随机取5只仔鼠,灌流固定大脑,进行组织病理切片和免疫组化染色,观察分析内嗅皮质CaMKⅡ表达。结果PN7,PN14和PN21时,15 ppm组和碘缺乏组仔鼠内嗅皮质CaMKⅡ的表达显著低于对照组(P<0.05),PN28和PN42时,5 ppm组、15 ppm组和碘缺乏组仔鼠内嗅皮质CaMKⅡ的表达亦显著低于对照组(P<0.05)。结论碘缺乏和甲状腺功能减退可降低内嗅皮质CaMKⅡ表达。
Objective To investigate the effects of entorhinal cortex and iodine deficiency and hypothyroidism on the expression of calcium and calmodulin-dependent kinase (CaMKⅡ). Methods Healthy 2-month-old female Wistar rats were randomly divided into control group, hypothyroidism group and iodine deficiency group according to their body weight. The hypothyroidism group was treated with propylthiouracil propylthiouracil (PTU) was divided into 5 ppm and 15 ppm groups, with 7 pregnant rats in each group. At PN7, PN14, PN21, PN28 and PN42 respectively, 5 pups were randomly selected from each group. Peripheral blood was collected and fixed for histopathology and immunohistochemistry. CaMKII expression in entorhinal cortex was observed and analyzed. Results The expression of CaMKII in the olfactory cortex of 15 ppm and iodine deficient groups was significantly lower than that of the control group (P <0.05) at PN7, PN14 and PN21. At PN28 and PN42, 5 ppm, 15 ppm and iodine deficiency groups The CaMKⅡexpression in entorhinal cortex of offspring rats was also significantly lower than that of the control group (P <0.05). Conclusion Iodine deficiency and hypothyroidism can reduce the expression of CaMK Ⅱ in entorhinal cortex.
