以陆地棉河南79的下胚轴为外植体,研究不同光质和凝固剂对愈伤组织诱导和生长的影响。试验结果表明,在光照培养条件下,外植体的出愈率为82.61%~98.36%,愈伤组织的生长较快,其鲜重和干重也较高。在黑暗和红光条件下,外植体易诱导产生不定根,外植体长根的百分率达到23.91%~54.10%,在不同凝固剂中以0.2%Gelrite对愈伤组织的诱导和生长效果为最佳。
The hypocotyls of Henan 79 were used as explants to study the effects of different light quality and coagulant on the induction and growth of callus. The results showed that under the light culture conditions, the explants’ curative rate was 82.61% ~ 98.36%, the callus grew rapidly, and the fresh weight and dry weight were also higher. Under dark and red light conditions, explants were easily induced to produce adventitious roots. The percentage of long roots of explants reached 23.91% ~ 54.10%. The effects of 0.2% Gelrite on callus Induction and growth effect is the best.
