目的:研究IFNγ对滑膜细胞分泌TNFα的影响。方法:将滑膜用胶原酶消化分离后获得的原代单个滑膜细胞(DSSC),加入不同剂量的IFNγ,观察脂多糖(LPS)存在或不存在下,不同时间(24小时、48小时、72小时)滑膜细胞分泌TNFα的情况;并观察了IFNγ对培养的滑膜纤维母细胞样细胞(SFC)在48小时时分泌TNFα的影响。用ELISA双抗体夹心法来检测培养细胞上清液中TNFα的含量,用配对t检验来统计结果。结果:不论LPS存在与否,IFNγ对RA和OA滑膜DSSC细胞分泌TNFα无明显的影响,但IFNγ对RA的SFC分泌TNFα有抑制作用(48小时)。结论:IFNγ对纤维母细胞分泌TNFα有抑制作用
Objective: To study the effects of IFN-γ on TNF-α secreted by synovial cells. Methods: Primary synoviocytes (DSSCs) obtained after collagenase digestion of synovial membrane were added with different dosages of IFNγ to observe the effect of lipopolysaccharide (LPS) in different time (24h, 48h) Hour, 72 hours) of synovial cells secreting TNF α situation; and observed IFN of cultured synovial fibroblast-like cells (SFC) at 48 hours to secrete TNF α. ELISA double antibody sandwich method to detect the culture supernatant of TNF α content, using the paired t test to the statistical results. Results: Regardless of the presence or absence of LPS, IFN γ had no significant effect on the secretion of TNFα by DSSC cells from RA and OA synovial cells, but IFNγ inhibited the secretion of TNFα by SFC (48 hours).
