目的:建立以反相高效液相色谱法同时测定罗哌卡因-醋酸地塞米松聚乳酸羟基乙酸共聚物(PLGA)微球中罗哌卡因和醋酸地塞米松含量的方法。方法:色谱柱为C_(18),流动相为乙腈-甲醇-20 mmol·L~(-1)磷酸二氢钠缓冲液(26:18:56),流速为1 mL·min~(-1),进样量为20μL,罗哌卡因和醋酸地塞米松检测波长分别为210、240 nm。结果:罗哌卡因和醋酸地塞米松检测浓度的线性范围分别为0.2~10、0.2~16μg·mL~(-1)(r=0.999 9);加样回收率均大于97.50%,RSD均小于2.59%。结论:本方法操作简便、重现性好、结果准确,可用于该制剂的含量测定。
Objective: To establish a method for simultaneous determination of ropivacaine and dexamethasone acetate in ropivacaine-dexamethasone (PLGA) microspheres by reversed-phase high performance liquid chromatography. Methods: The mobile phase consisted of acetonitrile and methanol (20 mmol·L -1) monobasic sodium phosphate buffer (26:18:56) at a flow rate of 1 mL · min -1 ), Injection volume of 20μL, ropivacaine and dexamethasone acetate detection wavelength was 210,240 nm. Results: The linear ranges of ropivacaine and dexamethasone acetate were 0.2 ~ 10 and 0.2 ~ 16 μg · mL -1, respectively (r = 0.999 9), and the recoveries were both higher than 97.50% Less than 2.59%. Conclusion: The method is simple, reproducible, accurate and can be used for the determination of the content of the preparation.
