目的研究哮喘大鼠肺组织中白三烯B_4(LTB_4)、白三烯C_4(LTC_4)及白三烯D_4(LTD_4)含量的变化及中药复方双龙胶囊对其的影响。方法将SD大鼠随机分成空白组、哮喘模型组、对照组(地塞米松组)、高、中、低3个中药剂量组。用卵蛋白致敏大鼠,以抗原攻击后建立哮喘模型,并用反向高效液相色谱法(RP-HPLC)测定各组大鼠肺组织匀浆中LTB_4、LTC_4、LTD_4含量,并进行比较。结果哮喘大鼠肺组织中LTB_4、LTC_4、LTD_4含量较正常对照组比均有显著升高,地塞米松和双龙胶囊8.27 g/kg,4.13 g/kg均可明显抑制哮喘大鼠肺组织中白三烯的生成(P<0.05)。结论双龙胶囊可明显抑制哮喘大鼠肺组织中炎性介质白三烯的生成,这可能是双龙胶囊用于治疗支气管哮喘的抗炎平喘作用机制之一。
Objective To investigate the changes of leukotriene B_4 (LTB_4), leukotriene C_4 (LTC_4) and leukotriene D_4 (LTD_4) in lung tissue of asthmatic rats and the effects of Shuanglong capsules. Methods SD rats were randomly divided into blank group, asthma model group, control group (dexamethasone group), high, middle and low doses of traditional Chinese medicine. Rats were sensitized with ovalbumin and challenged with antigen to establish an asthma model. The contents of LTB_4, LTC_4 and LTD_4 in lung homogenates of rats in each group were determined by reversed-phase high performance liquid chromatography (RP-HPLC) and compared. Results The contents of LTB_4, LTC_4 and LTD_4 in the lung tissue of asthmatic rats were significantly higher than those in the normal control group. Dexamethasone and Shuanglong Capsules 8.27 g/kg and 4.13 g/kg could significantly inhibit asthmatic rats. The production of leukotrienes in lung tissue (P<0.05). Conclusion Shuanglong Capsule can significantly inhibit the formation of inflammatory mediator leukotrienes in the lung tissue of asthmatic rats. This may be one of the anti-inflammatory and antiasthmatic mechanisms of Shuanglong Capsules for the treatment of bronchial asthma.
