200856 肺腺癌细胞系耐药及凋亡相关基因的表达和逆转/王洁…//中华肿瘤杂志.-1999,21(6).-422~420 将人工合成的反义、正义硫代脱氧寡核苷酸(S-ODM)经脂质体包裹后转染A_(549)~(DDP)细胞,用RT-PCR检测耐药及凋亡相关基因mRNA水平,免疫细胞化学及流式细胞-抗体(FCM-12)检测相应蛋白及核增殖抗原Ki-67表达,MTT检测其耐药性改变,TUNEL、DNA电泳检测细胞凋亡。结果,A_(549)~(DDP)细胞多药耐药基因(MDR1)、柘扑异构酶Ⅱ(TOPOⅡ)、c-myc mRNA及蛋白表达水平接近A_(549),二者c-erbB-2均阴性。A_(549)~(DDP)bcl-2阳性,且MRP mRNA及蛋白水平较

The antibody (FCM-12) was used to detect the expression of the corresponding protein and nuclear proliferation antigen Ki-67. MTT was used to detect the change of drug resistance. TUNEL and DNA electrophoresis were used to detect cell apoptosis. As a result, the levels of MDR1, TOPOII, c-myc mRNA and protein in A_(549)~(DDP) cells were close to that of A_(549), both c-erbB- 2 were negative.
