为了研究肿瘤转移的异质性,本实验通过使用单细胞克隆化技术,从体外培养的Lewis肺癌细胞中选出B、f 两亚细胞株,分别对它们进行扩大培养,再将其注射于小鼠足掌,发现B 细胞株的致瘤性和肺自发转移结节数均显著高于f 细胞株。进一步观察两株细胞其它方面的生物学特性发现:B细胞株体积大,粘附性差,呈铺展性生长。f 细胞株体积小,粘附性强。且B细胞株和较低浓度的ConA 发生凝集反应,而f 细胞株则在较高浓度ConA 中才发生凝集反应。这一结果提示:肿瘤细胞是由生物学特性不同的细胞亚群组成。
In order to study the heterogeneity of tumor metastasis, the two cell lines B and f were selected from in vitro cultured Lewis lung cancer cells using a single-cell cloning technique. They were expanded and cultured separately, and injected into smaller cells. Rat paws found that the tumorigenicity of the B cell line and the number of spontaneous metastatic lung nodules were significantly higher than those of the f cell line. Further observation of the biological characteristics of the other two cells revealed that: B cell lines are bulky and poorly adherent, showing spreading growth. f The cell strain is small and has strong adhesion. In addition, agglutination occurred in B cells and ConA at lower concentrations, whereas agglutination occurred in F cells at higher concentrations of ConA. This result suggests that the tumor cells are composed of subpopulations of cells with different biological characteristics.
